HOECHST 33342 / RHODAMINE 123 Mitochondrial Fluorescent Stain
This veterinary product combines two well-known fluorescent dyes—Hoechst 33342 and Rhodamine 123—for dual staining applications, particularly useful in assessing mitochondrial activity alongside nuclear visualization. Hoechst 33342 is a cell-permeable DNA stain that emits blue fluorescence and preferentially binds to adenine-thymine-rich regions in the minor groove of double-stranded DNA, suitable for both live and fixed cells. Rhodamine 123 is a cationic, cell-permeable fluorescent dye that rapidly accumulates in active mitochondria, emitting green to yellow-green fluorescence depending on the filter set used.

Main Benefits

Enables simultaneous visualization of nuclei (in blue) and active mitochondria (in green/yellow-green)
Hoechst 33342 effectively stains DNA in live or fixed cells without affecting cell permeability
Rhodamine 123 selectively accumulates in active mitochondria, allowing for mitochondrial function assessment
Rapid uptake of Rhodamine 123 facilitates efficient staining workflow
Compatible with fluorescence microscopy and flow cytometry applications

Composition

Hoechst 33342 (bis-benzimidazole DNA stain)
Rhodamine 123 (fluorescent dye sensitive to mitochondrial membrane potential)

Usage Instructions

Apply via fluorescent staining protocols using appropriate excitation/emission filters for Hoechst 33342 and Rhodamine 123
Hoechst 33342 binds to nuclear DNA, while Rhodamine 123 accumulates in active mitochondria
Dosage and incubation time should be determined by a qualified veterinarian or laboratory specialist based on species, sample type, and experimental conditions

Package Size

Typically offered as a dual staining kit or consumable set designed for mitochondrial activity assays (e.g., reference 15407/0012 in AndroVision® systems)

Note

Ideal for applications such as assessing mitochondrial activity in sperm, characterizing hematopoietic stem cells, or other research contexts requiring simultaneous visualization of nuclei and mitochondria
